Empirical evidence and theoretical understanding of ecosystem carbon and nitrogen cycle interactions

05 Nov 2024  |  nph.onlinelibrary.wiley.com
The article explores the interactions between carbon and nitrogen cycles in terrestrial ecosystems, emphasizing the role of nitrogen in limiting carbon cycle dynamics and the implications for climate change projections. It reviews the evolution of Dynamic Global Vegetation Models (DGVMs) from C-only to C-N models, highlighting the challenges and uncertainties in simulating carbon sinks and the influence of nitrogen availability. The text discusses empirical evidence from ecosystem experiments and the application of eco-evolutionary optimality (EEO) theory to improve model predictions. It underscores the need for better representation of C-N interactions in models to enhance the accuracy of climate projections.

A bioactive soluble recombinant mouse LIGHT promotes effective tumor immune cell infiltration delaying tumor growth

30 Jul 2024  |  www.biorxiv.org
The study explores the potential of a bioactive soluble recombinant mouse LIGHT protein, specifically Ig.Foldon-mLIGHT, in promoting immune cell infiltration and delaying tumor growth in a melanoma model. The research highlights the protein's ability to recruit cytotoxic and dendritic cells to the tumor site, suggesting its potential as a single agent or in combination with immune checkpoint blockade therapies to inhibit tumor progression.

A Universal, Continuous Assay for SAM-dependent Methyltransferases

01 Oct 2023  |  Wiley Online Library
The article discusses the development of a universal, continuous, and cell lysate-compatible assay for SAM-dependent methyltransferases. This assay is based on the enzymatic conversion of SAH to hydrogen sulfide, which can be fluorometrically detected in crude cell lysates. The assay addresses the limitations of current high-throughput screening methods for methyltransferase activity, such as the need for purified proteins and high background signals. The new assay demonstrates high sensitivity and selectivity, making it suitable for screening a wide range of methyltransferase variants and substrates. The study also highlights the potential applications of this assay in drug development and enzyme engineering.

A Causal Treatment for X-Linked Hypohidrotic Ectodermal Dysplasia: Long-Term Results of Short-Term Perinatal Ectodysplasin A1 Replacement

12 Apr 2023  |  mdpi.com
X-linked hypohidrotic ectodermal dysplasia (XLHED), a rare genetic disorder, has been treated successfully with a recombinant ectodysplasin A1 replacement protein, Fc-EDA. The treatment was administered either postnatally or prenatally at 26 weeks of pregnancy. Long-term follow-up showed that prenatal treatment led to the development of sweat glands and the ability to sweat, which are typically absent in untreated individuals with XLHED. The study supports further clinical investigation of EDA1 replacement therapy in utero as a new means of treating genetic disabilities.
